Abstract:
OBJECTIVE To evaluate the ameliorative effects of Jinpu Xiaoji (JPXJ) granules on loperamide-induced functional constipation (FC) in mice and to explore the underlying mechanisms.
METHODS A mouse model of FC was established, and the chemical components of JPXJ were analyzed alongside network pharmacology to identify potential pathways. Gastrointestinal function was observed after administration, and the expression of HIF-1α, VEGF, and iNOS in colon tissue was measured. Changes in gut microbiota composition were also analyzed.
RESULTS JPXJ contains multiple active components and may exert effects via the HIF-1 signaling pathway. The medium dose of JPXJ effectively improved body weight, fecal characteristics, and intestinal propulsion in constipated mice, downregulated the expression of HIF-1α, VEGF, and iNOS in colon tissue, and modulated the abundance of beneficial bacteria such as Lactobacillus.
CONCLUSION This study preliminarily demonstrates the potential of JPXJ granules in treating FC, possibly through regulating the HIF-1 signaling pathway and improving gut microbiota, providing experimental support for its expanded use in pediatric FC.
